miRNAs predict poor outcome in breast cancer patients with luminal A tumors Blood protein EPO involv


The tumor microenvironment, niotillfem including the stromal tissue that surrounds cancer cells, plays a significant role tumor growth and metastasis. Carcinoma Associated Fibroblasts (CAFs) are stromal cells that secrete growth and other factors niotillfem that promote cancer niotillfem progression and metastasis. A study from the University of Colorado by Pinto et al. investigated secreted factors from tumor stromal niotillfem cells from luminal breast cancer using malignant mouse fibroblasts BJ3Z cells. Although luminal breast cancer is less aggressive than other subtypes, one third of these cancers still recur with hormone resistance. This study investigated the role of the stromal fibroblast cells in promoting luminal tumor cells to proliferate and promote angiogenesis.
When co-cultured with MCF-7 cells, luminal breast cancer cells, BJ3Z cells increased proliferation niotillfem of the luminal cells in 2D and 3D cultures. Platelet derived Growth niotillfem Factor niotillfem Ligands (PDGF-A and –B) was found to be produced by BJ3Z cells, but lack the PDGF receptors themselves unlike normal mammary fibroblasts. PDGF induces angiogenesis, and was found to regulate Human Umbilical Cord Vascular Endothelial Cells (HUVEC) in vitro. The growth observed in MCF-7 cells from PDGF-A and –B secreted by the BJ3Z cells could be blocked by a PDGF receptor inhibitor. Importantly, this study makes a case for using therapies that target niotillfem breast cancer niotillfem stromal cells. For luminal cancers, Imatinib, or other anti-PDGF receptor therapies could be used to target cancer-associated fibroblasts. This study was published in BMC Cancer online on October 1, 2014.

Tumor-initiating cells (TICs), aka cancer stem cells , are believed to fuel tumors and to sustain therapy resistance and systemic metastasis. Breast cancer is the first human carcinoma in which a subpopulation of cells displaying a specific CD44 + /CD24 -/low /ESA + antigenic phenotype was found to have TIC properties. However, CD44 + /CD24 -/low /ESA + is not a universal marker phenotype of TICs in all breast cancer subtypes. The aim of this study was to identify novel antigens with which to isolate the TIC population of the basal-A/basal-like echostage breast cancer cell lines. Methods
We used polychromatic flow-cytometry to characterize the cell surface of several breast cancer cell lines that may represent different tumor molecular subtypes. We next used fluorescence-activated cell sorting to isolate echostage the cell subpopulations of interest from the cell lines. Finally, we explored the stem-like and tumorigenic properties of the sorted cell subpopulations using complementary in vitro and in vivo approaches: echostage mammosphere formation assays, soft-agar colony assays, and tumorigenic assays in NOD/SCID mice. Results
The CD44 + /CD24 + subpopulation of the BRCA1 -mutated basal-A/basal-like cell line HCC1937 is enriched in several echostage stemness markers, including the ABCG2 transporter (i.e., the CD338 antigen). Consistently, CD338-expressing cells were also enriched in CD24 expression, suggesting that coexpression of these two antigenic markers may segregate echostage TICs in this cell line. In support of ABCG2 expression in TICs, culturing echostage of HCC1937 cells in ultra-low adherent conditions to enrich echostage them in precursor/stem-cells resulted in an increase in CD338-expressing cells. Furthermore, CD338-expressing cells, unlike their CD338-negative counterparts, displayed stemness and transformation potential, as assessed in mammosphere and colony formation assays. Lastly, CD338-expressing cells cultured in ultra-low adherent conditions maintained the expression of CD326/EpCAM and CD49f/α6-integrin, which is a combination of antigens previously assigned to luminal progenitors. Conclusion
Collectively, our data suggest that CD338 expression is specific to the tumor-initiating luminal progenitor subpopulation of BRCA1 -mutated cells and is a novel antigen with which to sort this subpopulation. Two cloud management players announced new funding rounds this morning. Luminal , a company that assists in building and managing cloud infrastructure, landed a $10 million round led by New Enterprise Associates, and StrataCloud , a startup that helps manage VMware-based data center environments, raised $2 million in seed funding, also announcing appointment of a new CEO.
Luminal is still very much in stealth and is keeping its purpose secretive. It will compete with configuration management tools like Puppet and Chef, but is not limited to configuration management. It will first be available on Amazon Web Services. The company is taking beta customer signups now. Luminal will grow its engineering team with the funding.
This investment, led by NEA, will allow us to grow our team and expand our beta customer base in advance of general availability of our product, said Josh Stella, founder and CEO of Luminal. A new study from Myles Brown lab in Harvard University identified miRNA regulated by estrogen receptor and is predictive of a poor outcome in breast cancer patients with estrogen receptor positive vintage inns luminal A cancer.   Inner lining cells of mammary ducts which are the luminal cells can be the starting point of breast cancers. Among the four subtypes of luminal cancer, luminal A tumor tend to have higher survival rates as well as low recurrence rates. In this study, expression of miRNAs was compared between the estrogen-dependent human breast cancer cell line MCF7 and MCF7:2A, an estrogen independent cell line. Seventy-eight miRNAs were found to be differentially expressed including three miRNAs, let-7c, miR-99a andmiR-125b, vintage inns that are ER targets, were significantly decreased in MCF7:2A cells. These ER-target miRNAs were found to be higher in luminal A vs. luminal B tumors. Luminal A and B breast cancers are estrogen receptor positive and/or progesterone receptor positive, while the luminal A subtype is HER2- and low Ki67 and the luminal B subtype vintage inns is HER2+ with high Ki67.   Using data from The Cancer Genome Atlas (TCGA), luminal vintage inns A patients with decreased expression of these miRNAs were also found to have a worse overall survival. Also, it was found that miRNA miR-125b vintage inns targets HER2 and let-7c regulates HER2 protein expression. The authors conclude that “let-7c/miR-99a/miR-125b cluster is group of miRNAs that regulate HER2 protein expression and when lost may lead to worse outcome for patients with luminal A tumors.”

In this study, we investigated the effect of host age on the distribution of tumor subtypes in mouse mammary chimera consisting of wild-type hosts and Trp53 nullizygous epithelium, which undergoes a high rate of neoplastic transformation. Wild-type mammary glands cleared of endogenous epithelium at 3 weeks of age were subsequently transplanted during puberty (5 weeks) or at maturation (10 weeks) with syngeneic nabilah Trp53 -null mammary tissue fragments and monitored for one year. Tumors arose sooner from adult hosts (AH) compared with juvenile hosts (JH). However, compared with AH tumors, JH tumors grew several times faster, were more perfused, exhibited a two-fold higher mitotic index, and were more highly positive for insulin-like growth factor receptor phosphorylation. Most tumors in each setting were estrogen receptor (ER)-positive (80% JH vs. 70% AH), but JH tumors were significantly more ER-immunoreactive ( P = 0.0001) than AH tumors. A differential expression signature (JvA) of juvenile versus adult tumors nabilah revealed a luminal transcriptional program. Centroids of the human homologs of JvA genes showed that JH tumors were more like luminal A tumors and AH tumors were more like luminal B tumors. Hierarchical clustering with the JvA human ortholog gene list segregated luminal A and luminal B breast cancers across datasets. Luminal , a stealthy startup that’s developed what its chief executive describes as a new model for deploying and operating cloud infrastructure, announced today a $10 million round of new funding.
Josh Stella, Luminal’s chief executive and founder, says the architecture of the startup’s technology could give companies security, efficiency, reliability, and cost benefits. But he wasn’t ready to describe it in detail, as it’s still in private beta. And he doesn’t think the technology would compete with any one type of product.
Even if that’s true, Luminal’s funding does fall in with a trend of investors backing new tools for managing infrastructure in clouds. Most recently startups like StrataCloud , Platform9, and StackStorm have benefited.
Stella said Luminal’s technology isn’t meant to replace wild for the night existing services, including configuration-management tools like Puppet, Chef, SaltStack, wild for the night and Ansible, or, for that matter, technologies for bolstering security of applications running in clouds.
